El CReSA participarà en els propers esdeveniments europeus sobre Immunologia

Investigadors del CReSA participaran amb un total de vuit comunicacions en el 3r EVIW (Taller Europeu d’Immunologia Veterinària, 10-13 Setembre 2009) i en el 2n ECI (Congrés Europeu d’Immunologia, 13-16 Setembre 2009), que es celebraran a Berlín, Alemanya.

El programa científic del 2n ECI cobrirà els desenvolupaments més recents en immunologia bàsica i clínica, mentre que el 3r EVIW inclourà els següents temes:

  • Poblacions de limfòcits i regulació immune
  • Citoquines i quimioquines

CHARACTERIZATION OF IL-10 PRODUCING CELLS IN SPLEEN OF NATURAL CASES OF POSTWEANING MULTISYSTEMIC WASTING SYNDROME (PMWS) IN PIGS

The main objective of this work was to characterize and localize different immune cell subpopulations involved in IL-10 secretion in the spleen of pigs with PMWS using an immunofluorescence assay (IFA). This is the first immunophenotyping study on IL-10 producing cells in spleen of natural cases of PMWS. The present study have shown that macrophages and/or monocytes (CD163+ cells) and T lymphocytes (CD4+cells, CD8+ cells) produce IL-10 and further extend previous in vitro and in vivo studies on the role of IL-10 in PMWS pathogenesis.

CALICIVIRUS-LIKE PARTICLES MODULATE PORCINE DENDRITIC CELLS IN VITRO

We have analyzed the immunogenic potential of virus-like particles (VLPs) from rabbit hemorrhagic disease virus (RHDV) in porcine bone marrow-derived dendritic cells (poBMDCs), as we have previously described in mice. Our data showed that the RHDV-VLPs modulate the poBMDCs response and show their immunogenic potential for new vaccine development against animal and human viral infections.

CHIMERIC CALICIVIRUS-LIKE PARTICLES ELICIT PROTECTIVE ANTI-VIRAL CYTOTOXIC RESPONSES WITHOUT ADJUVANT

We have analyzed the potential of virus-like particles (VLPs) from rabbit hemorrhagic disease virus (RHDV) as a delivery system for foreign T-cell epitopes. Our data demonstrated that immunization with chimeric RHDV VLPs was able to protect mice from a viral challenge, suggesting the potential suitability of these constructions for new vaccine development against animal and human viral infections.

DIFFERENT IMMUNOREGULATORY EFFECT OF PORCINE REPRODUCTIVE AND RESPIRATORY VIRUS STRAINS ON PORCINE MYELOID DENDRITIC CELLS

The present study examines the effect of three European-type strains (3262, 3267 and 3249) representative of different patterns of cytokine induction in BMDC (IL-10+/TNF-a+ for 3262; IL-10-/TNF-a+ for 3249, and no cytokine release for 3267) on cell markers. In summary, different PRRSV strains have different abilities to regulate the expression of key molecules involved in antigen presentation and this seems to correlate with the cytokine profiles induced in BM-DC.

In the present study, 7 PRRSV European-type strains were fully sequenced and the amino acid sequences were predicted. Taking together the present results suggest that understanding of the humoral response against PRRSV should be approached considering the effects of this genetic diversity upon B cell epitopes.

SWINE INFLUENZA VIRUS (H3N2) INFECTION AND POLY-IC STIMULATION DIFFERENTIALLY UP-REGULATE SURFACE MARKERS AND CYTOKINE SECRETION ON PORCINE DENDRITIC CELLS

Our main goal was to characterise dendritic cell (DC) interaction with swine influenza virus (SIV) in vitro. Ultrastructural and phenotypical analysis of our DC corresponds with previously described data. DC exhibited a differential response to TLR agonists, suggesting a specific function for pathogen detection. Up-regulation of surface markers and increased levels of innate cytokines after SIV infection in vitro correlate with data from an early SIV infection in vivo. These data pave the way for understanding the intimate relation between SIV and porcine dendritic cells for triggering a protective immune response.
